There is no reply-to address in smtp. smtp defines an envelope sender (used with the MAIL FROM command). this addres must be valid. mail from invalid envelope senders can safely be rejected. That said, don't use address verification without being authorized by the owner of the sender domain. unauthorized probes are a form of abuse.
