I'm setting followups to the mozilla.support.thunderbird newsgroup,
which is more appropriate for this question.

You haven't yet set up a newsgroup account, have you? On Windows, click
"Tools -> Account Settings..." then (after checking that there isn't yet
an account for the server you want to use), click the "New" button near
bottom left. You'll be taken through several pages of settings, after
what, if they are set up correctly, you ought to be able to subscribe to
the newsgroups available at that server.

Alternately, if you did create a newsgroup account, then maybe one or
more of its settings are in error? Open the Account Settings just like
above, then click successively on the server name (in the left pane) and
on each of its subheadings, and check all the settings in those various
settings pages. A single typo in the server address (such as
news.mizilla.org instead of news.mozilla.org, or smnews.microsoft.com
instead of msnews.microsoft.com) can be enough to prevent you from
connecting, so double-check everything.

If you find and correct the error, you ought to be able to subscribe to
newsgoups from the main (three-pane) window, either by left-clicking the
server name in the left pane, then "Manage Subscriptions" in the right
one, or by right-clicking the server name, then Subscribe...
